Purpose
To compare subfoveal choroidal thickness (SCT) obtained from the topographic map of choroidal thickness in ETDRS-style grid and the manual line measurement in healthy eyes and to evaluate the influence of several ocular parameters over SCT.

Methods
Prospective and non-randomized study with 40 eyes of 40 healthy volunteers aged between 18 and 45 years. SCT was automatically obtained from ETDRS grid using the 12mm line scan pattern. Then, SCT was manually measured as the distance between the posterior edge of the hyper-reflective retinal pigment epithelium (RPE) and the choroidal junction using the single line scan pattern. Both SCTs were measured using swept-source OCT (DRI-1 Atlantis, Topcon Medical System, Japan). Only eyes with axial length (AL) between 22-26mm and spherical equivalent (SE)<-3.00D were included. The t-test was used to compare SCT between the two scanning methods. Multiple linear regression was performed to evaluate the influence of gender, age, SE and AL over SCT.
Results
Mean age was 29.85 ± 7.10years, mean SE was -0.70 ± 1.31D and mean AL was 23.52 ± 0.92mm. SCT was thinner at central ring, 309 ± 62μm, compared to the manual SCT measurement, 335 ± 66μm. The two different scanning protocols showed significatively differences, mean difference= 25.55 ± 27.07μm (95%CI, 16.90-34.21), t(39)=5.97, p=0.000. Similar SCTs were observed between women and men at the central subfield of the choroidal thickness map (women 335 ± 62μm vs. men 331 ± 84μm) and with manual measurement (women 311 ± 58μm vs. men 302 ± 80μm). Multiple linear regression showed that SCT was not significantly associated with either gender, age, SE or AL, although a trend for thinner SCT associated with longer AL was observed (B= -13.2μm), p>0.05.
Conclusions
Choroidal thickness measured by using ETDRS grid reported SCT significantly thinner than manual measurement. Both methods should be independently considered. SCT did not varied significantly with gender, age, SE and AL.
