Corneal histopathological changes due to topical mitomycin C application in different doses and periods in albino rabbits

Abstract Details

Purpose:

To evaluate the histopathological changes due to topical mitomycin C application in different doses and periods in albino rabbits.

Setting:

Cukurova University School of Medicine Department of Ophthalmology, Adana-TURKEY

Methods:

The study group consisted of 35 albino rabbits with a mean age of 6 months. The rabbits were divided into 7 groups each consisting of 10 eyes of 5 rabbits. Five rabbits were used as control. The first 3 groups underwent 0.2, 0.4, 0.8 mg/ml mitomycin C application 4x1 subsequently, 7 consecutive days for 6 cycle. Groups 4,5 and 6 underwent 0.2, 0.4, 0.8 mg/ml topical mitomycin C application 4x1, 7 consecutive days for 12 cycle. Control group underwent topical isotonic application. After the termination of applications, enucleation was performed and all the rabbits were sacrified with intracardiac ketamine injection.

Results:

Light microscopic and electron microscopic examination of mitomycin C applied subjects revealed histopathologic changes in cornea ephitelium and endothelium in concordance with dose and application period.

Conclusions:

Topical mitomycin C has corneal toxic effects related with dose and period and is known to be more toxic especially in increased doses. We aimed to determine the most effective dose and period without toxic effects to cornea in order to prevent side effects due to topical mitomycin C application.
