Comparative evaluation of cytotoxicity of antimicrobial eye drops

To compare in vitro the overall cytotoxic effects of six antibacterial fluoroquinolone eye drops

The overall cytotoxic effects of Cipromed™ (ciprofloxacin 0,3%; Sentiss Pharma Pvt. Ltd., India); Floxal™ (ofloxacin 0,3%; Dr. Gerhard Mann, Chem.-Pharm. Fabrik GmbH, Germany); Oftaquix™ (levofloxacin 0,5%; Santen Oy, Finland); Signicef® (levofloxacin 0,5%; Sentiss Pharma Pvt. Ltd., India); Vigamox® (moxifloxacin 0,5%; Alcon Laboratories, Inc., USA); and Zymar® (gatifloxacin 0,3%; Allergan Sales LLC, USA) were compared in vitro. Two types of cells were used in the experiment: cells transformed with a constant cell line (ovary cells Chinese hamster tumor) and normal human skin fibroblasts. The concentration of the test drugs was equal to 1.25% of the volume of culture medium


The cytotoxic effects of antibacterial fluoroquinolone on the cells transformed with a constant cell line were found in the following order (descending toxicity): moxifloxacin greater-than levofloxacin (original) greater-than levofloxacin (generic) greater-than gatifloxacin greater-than ciprofloxacin greater-than ofloxacin. The cytotoxic effects of antibacterial fluoroquinolone on the human skin fibroblasts were found in the following order (in descending toxicity): ciprofloxacin ≥ moxifloxacin, gatifloxacin greater-than levofloxacin (generic) = levofloxacin (original) greater-than ofloxacin


The study showed a possibility of using cultured cells for comparative evaluation of the cytotoxic effects of various ophthalmic preparations. We found that antimicrobial medicines that had been tested in vitro differ in their cytotoxic potential
