To determine the adherence of Rat Mesothelial cells sheets on a human devitalized lens capsule as a carrier.

Culture of Mesothelial Rat Cells were done and 34 Anterior Lens Capsules were obtained during normal cataract surgery procedures. In 6 cases the capsulorhexis was done with a femtosecond laser system achieving a size of 4.5mm. In the rest of the cases manual capsulorhexis was done targeting a 5mm diameter. All the capsules where stored immediately in distilled water in order to accomplish a total devitalized basal membrane. Seeding of Mesothelial Rat Cells in Anterior Lens Capsule was done and evaluation methods after staining was done with optic and electronic microscope.
Cell density was correctly achieved after the culture been in physiological levels. Cell morphology was similar to human corneal endothelial cells. Excellent adherence between rat mesothelial cells and basal membrane was observed, also cell-cell tight junction was assesed.
Similar morphology was found between Cultures of Endothelial Cells and Mesothelial Rat Cells. Excellent adhesion between cells and complex cell-basal membrane was accomplished. Density of Mesotelial Rat Cells achieved in culture and seeding were better than expected. Further animal experimenation need to be done with Human Mesothelial Cells in order to asses the functionality of these substitution for corneal endothelium.
