Abstract Details

Purpose:

To investigate the efficacy of linezolid, imipenem and vancomycin against biofilm that was produced by methicillin-resistant Staphylococcus spp.

Setting:

Anadolu University, Faculty of Science, Department of Microbiology

Methods:

Biofilm production was detected using microtiter assay. Staphylococcus spp. culture were diluted 1:100 and 200µl of the diluted cultures per well, were inoculated into 96well polystrene microtiter plates. Plates incubated for 3 days at 37oC. After incubation plates were poured and then 40µg/ml vancomycin or 40µg/ml imipenem or 8µg/ml linezolid was added into the different plates that biofilm had already formed; and incubated at 37oC for 24 hours. After incubation and broth removal, the plate was washed with sterile PBS stained with crystal violet. Optical density was measured at 570 nm using the microtiter plate reader.

Results:

We studied 10 S. epidermidis, 6 S. lugdunensis, 6 S. warneri, 2 S. saprophyticus, 1 S. hominis and 1 S. aureus isolates. Imipenem was the most effective antibiotic against 13 isolates, most of them were S. epidermidis (5 isolates) and S. warneri (4 isolates); linezolid was most effective antibiotic against 11 isolates, most of them were S. epidermidis (5 isolates) and S. lugdunensis (4 isolates). Vancomycin was most effective antibiotic against 2 isolates. Optical density (OD) value was 2.01±0.68 for control group. OD values for vancomycin, linezolid and imipenem were 1.32±0.71, 0.84±0.73, 0.78±0.67 respectively(p<0.001). Efficacy of linezolid and imipenem were similar.

Conclusions:

Our results suggested that effects of imipenem and linezolid against biofilm that formed by methicillin-resistant Staphylococcus spp. were greater than effect of vancomycin. All of the three antibiotics were significantly effective against biofilm of S. warneri isolates, linezolid was significantly effective against biofilm of S. lugdunensis isolates, imipenem was significantly effective against biofilm of S. epidermidis isolates.
